Arbuscular mycorrhizal fungi (AMF) are a key group of soil organisms involved in several ecosystem services, but they had not been explored in retrospective ecological risk assessment (ERA) schemes, partially due to a lack of experimental protocols. We aimed to perform a site-specific test to assess the effects of multiple metal contaminated soil (Zn, Cu, Cd, and Pb) on several ecotypes of AMF and evaluate the conceptual fitness of the performed AMF test to include in the TRIAD approach of ERA schemes. The results demonstrated that increasing metal contaminated soil proportions that inhibited 50 % (IC50) of spore germination varied from 28 to >80. Ecotypes such as Claroideoglomus etunicatum (IC50 > 80) and Racocetra gregaria (IC50 > 80) experienced 50 % reduction in spore germination at metals concentrations of 10,776.3 for Zn, 1015.2 Cu, 65.5 Cd, 140.2 mg dm-3 Pb, that are 3 times higher than those for Acaulospora mellea CMM101 (IC50 28 [16.2-39.8]) (3441.7 Zn, 333.9 Cu, 17.8 Cd, 56.5 mg dm-3 Pb). In the evaluation of the suitability of the AMF ecotoxicological test to ERA, both spore germination and germinative tube growth were best evaluated and thus suitable in the following descending order: Tier III, Tier II, and Tier I. Variable effects of multiple-metal contamination on the ecotypes indicates how AMF community is affected in its pre-symbiotic structures. The ecotoxicological test allowed the selection of two species with the greatest sensitivity (Ambispora appendicula and Rhizophagus clarus CMM103) to the metal matrix, with the potential to best fit ERA objectives. The site-specific ecotoxicological test with AMF ex-situ proved adequate as an alternative test for Tiers II and III of TRIAD ERA schemes for metal contaminated areas. Data generated through test results, such as the inhibition concentrations (ICs), could be incorporated into ERAs risk indexes, increasing its ecological relevance, and reducing overall uncertainties.

The objective of this study was to evaluate the agronomic efficiency of rhizobia strains already approved or in the selection stage for cowpea, in the Recôncavo da Bahia region, Brazil. Two field experiments were performed in the municipalities of Cruz das Almas and Maragogipe, Bahia, Brazil. A randomized block design was used with seven nitrogen sources and four replicates. Nitrogen sources consisted of the strains UFRB FA51B1, UFRB BA72C2-1, UFLA 03-164T, UFLA 03-84, INPA 03-11B, and two controls without inoculation, one with mineral nitrogen and another without. All the strains nodulated cowpea. The efficiency of the strains was determined by the number of nodules, nodule dry matter, total dry matter, grain yield, nitrogen accumulation in shoots and grain, and relative efficiency. The strain UFLA 03-164T can be recommended for biomass production, green manure, and promotion of grain yields in both soils. In Maragogipe, the UFLA 03-84 and INPA 03-11B strains can be recommended for biomass production, green manure, and promotion of grain yields. The UFLA 03-164T strain showed great potential to promote and grain yield in the two municipalities studied. The strain INPA 03-11B can be recommended for Maragogipe soil. UFLA 03-84 can also be recommended to increase grain yield in Cruz das Almas.

Novel green technologies for soil remediation have been focusing on altering soil properties and improving soil health. Hydrothermally-altered feldspar (HYP, HydroPotash), recently developed, is being related as both an efficient amendment to immobilize heavy metals in soils and a plant nutrients source, consisting in a promising technology for revegetation of contaminated sites. In order to evaluate the effectiveness of using HYP for phytostabilization programs, two different soils (Technosol and Oxisol) collected from a smelting site were amended with increasing doses of HYPs (HYP-1 and HYP-2): 15, 30, 60, and 120 Mg ha-1. For comparison, a control (soil without amendment) and a soil amended with zeolite (clinoptilolite) were also included as treatments. After 90 days of incubation, HYPs decreased up to 83.8 % of Cd availability and reduced exchangeable Al up to 100 %. HydroPotash increased pH, cation exchange capacity, and contents of potassium, calcium, and phosphorus, as well as microbial biomass carbon, and fluorescein diacetate hydrolysis of soils. Andropogon gayanus, Eucalyptus grandis, and Heterocondylus vitalbae started growing from the dose of 15 Mg ha-1 HYPs in the Oxisol and 60 Mg ha-1 HYPs in the Technosol. Principal component analysis indicates that plant shoot dry weight was negatively correlated with extractable Cd and Zn and positively with pH, CEC, and Ca content. Besides promoting plant growth, HYPs reduced heavy metals (Cd and Zn) absorption by plants, indicating that HYP has potential use as an amendment in phytostabilization programs.

The evaluation of the chemical leaching potential from soils amended with biosolid is of extreme importance for environmental safety of agricultural use of these residues. The objective of this study was to evaluate the polluting potential and possible risks associated with the agricultural use of biosolids generated by the polyethylene terephthalate (PET) fiber and resin industry through ionic speciation and analysis of the activity of chemical species present in the leached solution from Inceptisol treated with rates 0, 6, 12, 18, 24, 48, 96, and 144 Mg ha-1 on dry basis. The experiment was conducted in a lysimeter and the treatments with three replications were distributed at random. Chemical leaching was made by application of CaC12 0.01 mol L-1 solutions in a volume fourfold higher than the water retention capacity of the soil, divided into five leaching events: 210, 245, 280, 315, and 350 days of incubation. Chemical species concentrations in collected leachates were used for ionic speciation by geochemical software Visual MINTEQA2 version 4.0. Impact factor of chemical species was calculated as the ratio between maximum concentration in the leach solution in the treated soil and control. Dissolved organic carbon had strong influence on Pb+2 and Cu+2 leaching, but these elements in free or complexed forms presented low activities in solution. Leaching of NO3-, Zn+2, and Na+ represents the main environmental risk of agricultural use of this residue. However, these risks can be minimized if technical criteria and critical limits for the agronomic use of biosolids were observed.

Revegetation of areas with mine tailings should consider not only the best technique but also the choice of species adapted to this condition. This study aimed to analyze the occurrence of plant species, their Mn concentrations, and mycorrhizal colonization after implementation of two revegetation techniques (replacement of topsoil with natural seed bank and planting of seedlings) in an area of disposal of tailings from Mn processing compared to a native forest area as well as to a place that was not revegetated. Plant samples (at least three individuals/species) were collected from the understory at revegetated locations and forest. The established plant species and their Mn concentrations, mycorrhizal colonization, and forms of Mn in the soil were analyzed. The use of topsoil led to greater plant diversity. The high concentrations of Mn in the substrates did not affect the occurrence of vegetation in the understory and mycorrhizal colonization. The plant species established in the revegetated areas differed in relation to Mn concentration (471-27,842 mg kg-1 in leaves), Mn translocation factor (0.2-125.3) and mycorrhizal colonization rates (1-35%). Four potential Mn hyperaccumulators species were identified: Aparisthmium cordatum, Clidemia hirta, Socratea exorrhiza, and Vismia latifolia.

ABSTRACT Sophora tomentosa is a pantropical legume species with potential for recovery of areas degraded by salinization, and for stabilization of sand dunes. However, few studies on this species have been carried out, and none regarding its symbiotic relationship with beneficial soil microorganisms. Therefore, this study aimed to evaluate the diversity of nitrogen-fixing bacteria isolated from nodules of Sophora tomentosa, and to analyze the occurrence of colonization of arbuscular mycorrhizal fungi on the roots of this legume in seafront soil. Thus, seeds, root nodules, and soil from the rhizosphere of Sophora tomentosa were collected. From the soil samples, trap cultures with this species were established to extract spores and to evaluate arbuscular mycorhizal fungi colonization in legume roots, as well as to capture rhizobia. Rhizobia strains were isolated from nodules collected in the field or from the trap cultures. Representative isolates of the groups obtained in the similarity dendrogram, based on phenotypic characteristics, had their 16S rRNA genes sequenced. The legume species showed nodules with indeterminate growth, and reddish color, distributed throughout the root. Fifty-one strains of these nodules were isolated, of which 21 were classified in the genus Bacillus, Brevibacillus, Paenibacillus, Rhizobium and especially Sinorhizobium. Strains closely related to Sinorhizobium adhaerens were the predominant bacteria in nodules. The other genera found, with the exception of Rhizobium, are probably endophytic bacteria in the nodules. Arbuscular mycorrhizal fungi was observed colonizing the roots, but arbuscular mycorhizal fungi spores were not found in the trap cultures. Therefore Sophora tomentosa is associated with both arbuscular mycorhizal fungi and nodulating nitrogen-fixing bacteria.

ABSTRACT Determination of soil properties ​​helps in the correct management of soil fertility. The portable X-ray fluorescence spectrometer (pXRF) has been recently adopted to determine total chemical element contents in soils, allowing soil property inferences. However, these studies are still scarce in Brazil and other countries. The objectives of this work were to predict soil properties using pXRF data, comparing stepwise multiple linear regression (SMLR) and random forest (RF) methods, as well as mapping and validating soil properties. 120 soil samples were collected at three depths and submitted to laboratory analyses. pXRF was used in the samples and total element contents were determined. From pXRF data, SMLR and RF were used to predict soil laboratory results, reflecting soil properties, and the models were validated. The best method was used to spatialize soil properties. Using SMLR, models had high values of R² (≥0.8), however the highest accuracy was obtained in RF modeling. Exchangeable Ca, Al, Mg, potential and effective cation exchange capacity, soil organic matter, pH, and base saturation had adequate adjustment and accurate predictions with RF. Eight out of the 10 soil properties predicted by RF using pXRF data had CaO as the most important variable helping predictions, followed by P2O5, Zn and Cr. Maps generated using RF from pXRF data had high accuracy for six soil properties, reaching R2 up to 0.83. pXRF in association with RF can be used to predict soil properties with high accuracy at low cost and time, besides providing variables aiding digital soil mapping.


RESUMO A determinação de atributos do solo auxilia no correto manejo da sua fertilidade. O equipamento portátil de fluorescência de raios-X (pXRF) foi recentemente adotado para determinar o teor total de elementos químicos em solos, permitindo inferências sobre atributos do solo. No entanto, esses estudos ainda são escassos no Brasil e em outros países. Os objetivos deste trabalho foram prever atributos do solo a partir de dados do pXRF, comparando-se os métodos de regressão linear múltipla stepwise (SMLR) e de random forest (RF), além de mapear e validar atributos do solo. 120 amostras de solo foram coletadas em três profundidades e submetidas a análises laboratoriais. Utilizou-se o pXRF para leitura das amostras e determinou-se o teor total de elementos. A partir dos dados do pXRF, foram utilizadas SMLR e RF para predizer resultados laboratoriais, que refletem atributos do solo, e os modelos foram validados. O melhor método foi utilizado para espacializar os atributos do solo. Utilizando SMLR, os modelos apresentaram valores elevados de R² (≥0,8), porém maior acurácia foi obtida na modelagem com RF. A capacidade de troca de cátions potencial e efetiva, matéria orgânica do solo, pH, saturação por bases e teores trocáveis de Ca, Al e Mg apresentaram ajustes adequados e predições acuradas com RF. Dos dez atributos do solo preditos por RF a partir de dados do pXRF, sete apresentavam CaO como a variável mais importante para auxiliar as predições, seguido por P2O5, Zn e Cr. Os mapas gerados a partir de dados do pXRF usando RF apresentaram adequados valores de R² para seis atributos do solo, atingindo R2 de até 0,83. O pXRF em associação com RF pode ser usado para prever atributos do solo com elevada acurácia, com rapidez e a baixo custo, além de proporcionar variáveis que auxiliam o mapeamento digital de solos.

ABSTRACT Arbuscular mycorrhizal fungi (AMF) interact symbiotically with most plant species, facilitating revegetation of areas under rehabilitation. The aim of this study was to evaluate the inoculum potential, density, and diversity of AMF spores in five environments, as well as the relation of species with soil properties. Soil samples were collected in five environments in a mining area and its surroundings in the Quadrilátero Ferrífero, Minas Gerais (Brazil): tailings piles in rehabilitation with grass, canga, Cerrado, native forest, and eucalyptus plantation; these samples were subjected to chemical and physical analyses. Spores were directly extracted from field samples and from trap cultures (TCs) established in two locations in the Southeast and South regions of Brazil for taxonomic identification of the species. Species richness, the Shannon diversity index (H'), and equitability were determined. Principal component analysis (PCA) was used to identify soil properties that most influenced AMF occurrence. Spore density showed no significant difference among the environments. A total of 59 AMF species were found. This is the first report of the occurrence of Acaulospora nivalis and Acaulospora alpina in Brazil. Higher H' and species richness in the field were found in tailings piles and lower in canga. Canga showed higher inoculum potential. The development of TCs in two locations allowed a wider diversity of AMF species to be captured. Environments of the Quadrilátero Ferrífero are hotspots of AMF diversity, and the soil pH and exchangeable S and P contents are the properties that best explain the distribution of AMF species.


RESUMO Fungos micorrízicos arbusculares (AMF) interagem simbioticamente com a maioria das espécies de plantas, facilitando a revegetação de áreas sob reabilitação. O objetivo deste trabalho foi de avaliar o potencial de inóculo, densidade e diversidade de esporos de AMF em cinco ambientes, e a relação das espécies com atributos do solo. Amostras de solo foram coletadas em cinco ambientes em área de mineração e seu entorno no Quadrilátero Ferrífero, Minas Gerais (Brasil): pilha de rejeitos em reabilitação com capim, canga, Cerrado, mata nativa e plantação de eucalipto, e submetidas a análises química e física. A extração de esporos direto de amostras de campo e de culturas armadilha (TCs), estabelecidas em dois locais nas regiões Sul e Sudeste do Brasil, foi feita para identificação taxonômica das espécies. Foram determinados riqueza de espécies, índice de diversidade de Shannon (H') e equitabilidade. Análise de componentes principais (PCA) foi utilizada para identificar atributos do solo que mais influenciaram a ocorrência de AMF. A densidade de esporos não diferiu significativamente entre os ambientes. Um total de 59 espécies de AMF foram encontradas, destacando-se o primeiro relato de ocorrência de Acaulospora nivalis e Acaulospora alpina no Brasil. Maiores H' e riqueza de espécies no campo foram encontradas em pilhas de rejeitos e menores em canga, embora canga tenha apresentado maior potencial de inóculo. A condução de TCs em dois locais proporcionou captura de maior diversidade de espécies de AMF. Ambientes do Quadrilátero Ferrífero são hotspots de diversidade de AMF e o pH do solo e teores trocáveis ​​de S e P são os atributos que melhor explicam a distribuição das espécies de AMF.

To maximize the contribution of biological nitrogen fixation in common bean, it is necessary to use bacterial strains that are more adapted, competitive, and efficient in the symbiotic process. In this regard, the aim of this study was to evaluate the agronomic efficiency (AE) of three bacterial strains isolated from acid soils with high Al content from the Amazon region in an Argissolo Vermelho Distrófico típico soil (Typic Rhodustults - USDA Classification) from the municipality of Formiga, MG, Brazil. We compared their AE to that of the reference strain CIAT 899T and of two controls without inoculation (one without and another with 80 kg ha-1 of N-urea). The results indicated that inoculation with the strains UFLA 02-100 and UFLA 02-127 provides grain yield equivalent to inoculation with the reference strain and to the control with mineral N. Thus, both have potential for recommendation as inoculants for common bean.


Para maximizar a contribuição da fixação biológica de nitrogênio no feijoeiro-comum é necessária a utilização de estirpes de bactérias mais adaptadas, competitivas e eficientes no processo simbiótico. Nesse sentido, objetivou-se avaliar, em um Argissolo Vermelho Distrófico típico do município de Formiga-MG, a eficiência agronômica (EA) de três estirpes isoladas de solos ácidos e com alto teor de Al da Amazônia e comparar suas EA à da estirpe referência CIAT 899T e à de dois controles sem inoculação (um sem e outro com 80 kg ha-1 of N-ureia). Os resultados indicaram que a inoculação com as estirpes UFLA 02-100 e UFLA 02-127 propicia rendimento de grãos equivalente ao da estirpe referência e ao do controle com N mineral e que por isso, ambas têm potencial para ser recomendadas como inoculantes para o feijoeiro-comum.

A great number of species and individuals of scandent legumes establishing symbiosis with nitrogen fixing bacteria occurs in the Amazon Forest. These symbiosis probably play an important role in contributing to nitrogen incorporation in this ecossystem. The objectives of this study were to evaluate the growth of eight species of scandent legumes in five nursery substrates; to compare nodulation with rhizobia strains introduced or native to these substrates; and to characterize phenotypically and genetically these rhizobia. The experiment was carried out in a completely randomized design with five replications. Five to seven months after seedling emergency, according to the legume species, growth and nodulation parameters were determined. Rhizobia identification of strains was carried out by 16S rRNA gene partial sequencing. The survival of seedlings after the transplanting varied from 93 to 98%, in Ultisol (Argissolo in Brazilian classification), collected in primary forest, and fertilized with all nutrients, except nitrogen (ULTfert); and in a clay and sand mixture, in a ratio 3:2 (CONV), respectively. Species with height superior to 30 cm, in general, grew better in substrates with higher fertility: ULTfert and Humic Gley soil (HG). Seven out of the eight species were able to nodulate. The percentage of nodulation per substrate was: SAND, washed sand with mixed inoculum of 100 rhizobia strains plus fertilization (100), HG (80), CONV (100), ULT, A-horizon of red-yellow Ultisol collected in the Ducke Forest Reserve (Manaus) (44), and ULTfert (55%). Bradyrhizobium spp. were isolated from nodules of all species and substrates. Burkolderia fungorum was isolated from Dalbergia inundata. For Dalbergia riedelli and Dalbergia inundata, this is the first report on the identification of symbiotic strains. Um grande número de espécies e indivíduos de leguminosas escandentes estabelecendo simbiose com bactérias fixadoras de nitrogênio ocorrem na floresta Amazônica. Estas simbioses provavelmente desempenham importante papel, contribuindo para incorporação de nitrogênio neste ecossistema. Os objetivos deste trabalho foram avaliar o crescimento de oito espécies de leguminosas escandentes em cinco substratos no viveiro; comparar a nodulação com as estirpes de rizóbio introduzidas ou nativas destes substratos; e caracterizar esses rizóbios fenotípica e geneticamente. O experimento foi conduzido em delineamento inteiramente casualizado com cinco repetições. Cinco a sete meses após a emergência das mudas parâmetros de crescimento e nodulação foram determinados. A identificação das estirpes de rizóbios foi feita pelo sequenciamento parcial do gene 16S rRNA. A sobrevivência das mudas após o transplante variou de 93 a 98%, em um Argissolo (Ultisol de acordo com a classificação da USDA), coletado em floresta primária e adubado com todos os nutrientes, exceto nitrogênio (ULTfert), e na mistura argila e areia, na proporção 3:2 (CONV), respectivamente. Espécies que apresentaram altura superior a 30 cm, no geral, cresceram melhor nos substratos de maior fertilidade: ULTfert e solo Glei Húmico (GH). Das oito espécies estudadas, sete nodularam. A porcentagem de nodulação por substrato foi: AREIA, areia lavada com inóculo misto de 100 estirpes de rizóbio mais adubação (100), GH (80), CONV (100), ULT, horizonte A de Argissolo vermelho-amarelo coletado na Reserva Florestal Ducke (Manaus) (44) e ULTfert (55%). Bradyrhizobium spp. foram isolados de nódulos de todas as espécies e substratos. Burkolderia fungorum foi isolada de Dalbergia inundata. Este é o primeiro relato de identificação de estirpes simbióticas para Dalbergia riedelli e Dalbergia inundata. Anthropogenic pressures on tropical forests are rapidly intensifying, but our understanding of their implications for biological diversity is still very limited, especially with regard to soil biota, and in particular soil bacterial communities. Here we evaluated bacterial community composition and diversity across a gradient of land use intensity in the eastern Amazon from undisturbed primary forest, through primary forests varyingly disturbed by fire, regenerating secondary forest, pasture, and mechanized agriculture. Soil bacteria were assessed by paired-end Illumina sequencing of 16S rRNA gene fragments (V4 region). The resulting sequences were clustered into operational taxonomic units (OTU) at a 97% similarity threshold. Land use intensification increased the observed bacterial diversity (both OTU richness and community heterogeneity across space) and this effect was strongly associated with changes in soil pH. Moreover, land use intensification and subsequent changes in soil fertility, especially pH, altered the bacterial community composition, with pastures and areas of mechanized agriculture displaying the most contrasting communities in relation to undisturbed primary forest. Together, these results indicate that tropical forest conversion impacts soil bacteria not through loss of diversity, as previously thought, but mainly by imposing marked shifts on bacterial community composition, with unknown yet potentially important implications for ecological functions and services performed by these communities.

RESUMO

We report the 7.4-Mb draft genome sequence of Mesorhizobium sp. strain UFLA 01-765, a Gram-negative bacterium of the Phyllobacteriaceae isolated from Zn-mining soil in Minas Gerais, Brazil. This strain promotes plant growth, efficiently fixes N2 in symbiosis with Leucaena leucocephala on multicontaminated soil, and has potential for application in bioremediation of marginal lands.

Fixação simbiótica de nitrogênio pode ser uma alternativa para reduzir, ou mesmo substituir, a adubação nitrogenada do feijoeiro-comum, cultura na qual ainda há descrédito quanto aos benefícios da inoculação e que se caracteriza por aplicações de altas doses de N mineral. Com o objetivo de verificar a resposta de cultivares de feijoeiro-comum à inoculação das sementes com duas estirpes de rizóbio, foram conduzidos dois experimentos de campo na safra de inverno 2010 (Patos de Minas e Uberaba, MG). Utilizou-se o delineamento em blocos casualizados, com três repetições e esquema fatorial 8 x 3, envolvendo oito cultivares (União, Madrepérola, Supremo, Ouro Negro, Ouro Vermelho e Majestoso) e três tipos de inoculação (sementes não inoculadas e estirpes CIAT 899T e UFLA 04-173). Na floração, avaliaram-se o número e massa seca de nódulos, massa seca de parte aérea e teor e acúmulo de nitrogênio (N) na parte aérea e, na colheita, o rendimento de grãos com seus componentes primários, teor e acúmulo de N nos grãos. Constatou-se que as populações nativas promovem nodulação equivalente à das estirpes inoculadas. As estirpes CIAT 899T e UFLA 04-173 favorecem o crescimento vegetativo do feijoeiro. A estirpe CIAT 899T proporciona maior número de vagens e maior massa de grãos que a estirpe UFLA 04-173, e em Uberaba também proporciona maior rendimento de grãos. Quando a inoculação é realizada com a estirpe UFLA 04-173, as cultivares diferem quanto ao teor foliar de N. As cultivares Madrepérola, Ouro Negro e Ouro Vermelho apresentam mais nódulos e maior massa seca de parte aérea, além de maior produtividade em Patos de Minas. Ouro Negro situa-se entre as mais produtivas em Uberaba. Em Uberaba há maior acúmulo de matéria seca de nódulos e de parte aérea, mas o ambiente Patos de Minas resulta em maiores valores do rendimento de grãos e componentes.


The symbiotic nitrogen fixation can be an alternative to reduce, or even replace, the nitrogen (N) fertilization of the common bean crop, characterized by application of high doses of this nutrient. This work aimed to verify the response of common bean cultivars to the seed inoculation with two Rhizobium strains, conducting two field experiments in the winter-spring crop of 2010, at Patos de Minas and Uberaba, Minas Gerais State, Brazil. The experiment had a randomized block design, with three replications and a 8 x 3 factorial squemme, involving eight cultivars and three levels of inoculation. At flowering, it was evaluated the number of nodules, nodules dry mass, shoots dry weight, N content and accumulation in shoots and, at harvest, the grain yield and its primary components (100 grains weight, number of pods per plant, number of grains per pod) and the N content and accumulation in the grains. The Madrepérola, Ouro Negro and Ouro Vermelho cultivars present the highest number of nodules and higher dry mass; in Patos de Minas, the same cultivars show higher productivity, but in Uberaba, only Ouro Negro cultivar is among the most productive. In Uberaba there is the greater nodules dry weight and higher shoots dry matter, but the most favorable conditions in Patos de Minas results in higher values for grain yield and its components. In Uberaba, the CIAT 899T strain overcomes the UFLA 04-173 strain in relation to grain yield. In both environments, the CIAT 899T strain also shows superiority in number of pods per plant and weight of one hundred grains.

Forty-eight Burkholderia isolates from different land use systems in the Amazon region were compared to type strains of Burkholderia species for phenotypic and functional characteristics that can be used to promote plant growth. Most of these isolates (n=46) were obtained by using siratro (Macroptilium atropurpureum - 44) and common bean (Phaseolus vulgaris - 2) as the trap plant species; two isolates were obtained from nodules collected in the field from Indigofera suffruticosa and Pithecellobium sp. The evaluated characteristics were the following: colony characterisation on "79" medium, assimilation of different carbon sources, enzymatic activities, solubilisation of phosphates, nitrogenase activity and antifungal activity against Fusarium oxysporium f. sp. phaseoli. Whole cell protein profiles, 16S rRNA, gyrB, and recA gene sequencing and multilocus sequence typing were used to identify the isolates. The isolates showed different cultural and biochemical characteristics depending on the legume species from which they were obtained. Except for one isolate from I. suffruticosa, all isolates were able to solubilise calcium phosphate and present nitrogenase activity under free-living conditions. Only one isolate from common beans, showed antifungal activity. The forty four isolates from siratro nodules were identified as B. fungorum; isolates UFLA02-27 and UFLA02-28, obtained from common bean plants, were identified as B. contaminans; isolate INPA89A, isolated from Indigofera suffruticosa, was a close relative of B. caribensis but could not be assigned to an established species; isolate INPA42B, isolated from Pithecellobium sp., was identified as B. lata. This is the first report of nitrogenase activity in B. fungorum, B. lata and B. contaminans.

The Upper Solimões river region, western Amazon, is the homeland of indigenous populations and contains small-scale agricultural systems that are important for biodiversity conservation. Although traditional slash-and-burn agriculture is being practiced over many years, deforestation there is relatively small compared to other Amazon regions. Pastures are restricted to the vicinity of cities and do not spread to the small communities along the river. Inceptisols are the main soil order (>90%) in the area and have unique attributes including high Al content and high cation exchange capacity (CEC) due to the enrichment of the clay fraction with 2:1 secondary aluminosilicates. Despite its importance, few studies have focussed on this soil order when considering land use effects on the fertility of Amazon soils. Thus, the objective of this study was to evaluate changes in soil fertility of representative land use systems (LUSs) in the Upper Solimões region, namely: primary rainforest, old secondary forest, young secondary forest, agroforestry, pasture and agriculture. LUSs were significantly differentiated by the chemical attributes of their topsoil (0-20 cm). Secondary forests presented soil chemical attributes more similar to primary rainforest areas, while pastures exhibited the highest dissimilarity from all the other LUSs. As a whole, soil chemical changes among Inceptisols dominated LUSs showed patterns that were distinct from those reported from other Amazon soils like Oxisols and Ultisols. This is probably related to the presence of high-activity clays enriched in exchangeable aluminum that heavily influenced the soil chemical reactions over the expected importance of organic matter found in most studies conducted over Oxisol and Ultisol.

O presente trabalho objetivou avaliar a resposta de Phaseolus vulgaris (L.) cv. 'Talismã' à inoculação com quatro cepas isoladas da região da Amazônia, UFLA 02-100, UFLA 02-127, UFLA 02-86 e UFLA 02-68, comparadas à cepa CIAT-899 (SEMIA4077), autorizada pelo MAPA como inoculante para cultura do feijoeiro. O experimento foi conduzido em campo na Universidade Federal de Lavras, utilizando-se o delineamento experimental em DBC, com sete tratamentos e com seis repetições, sendo cinco estirpes citadas e dois controles não inoculados, um com N-mineral (80kg ha-1 de N) e outra sem N-mineral. Foram avaliados a nodulação (número e massa seca de nódulos), o crescimento (massa seca da parte aérea), o rendimento de grãos e o teor e acúmulo de nitrogênio na parte aérea e nos grãos, além da eficiência relativa. A cepa UFLA 02-68 se destacou e promoveu rendimento de grãos semelhante ao da testemunha com 80kg ha-1 de nitrogênio, superando as demais estirpes, inclusive a CIAT 889.


This paper aimed to evaluate the effect of inoculation on Phaseolus vulgaris (L.) cv. 'Talismã' with four rhizobia strains isolated from Amazon region: UFLA 02-100, UFLA 02-127, UFLA 02-86 and UFLA 02-68, compared to inoculation with strain CIAT-899, officialy authorized as inoculant to common beans by MAPA. The experiment was carried out in the 'Universidade Federal de Lavras' campus, in a randomized block design, with 6 replications and 7 treatments. Treatments were the five strains and two controls: one receiving mineral-N (80kg N ha-1) and the other an absolut control. Nodule number and dry matter weight, shoot dry matter weight, grain yields and N-content both in shoots and grain were evaluated. Inoculation with strain UFLA 02-68 promoted highest grain yields, similar to the control receiving mineral-N, their yields were even higher than that of strain CIAT 889.

Estudos de qualidade do solo são importantes para o entendimento da interação dessa com o ecossistema como um todo. Nesse contexto, os microrganismos do solo desempenham papel fundamental para manutenção e produtividade de vários agroecossistemas. Objetivou-se, neste trabalho avaliar os atributos biológicos (carbono da biomassa microbiana, respiração basal e quociente microbiano), além do teor de carbono orgânico, possíveis indicadores das alterações na qualidade do solo impostas pelos diferentes sistemas de manejo, em relação ao cerrado nativo, em sistema agrossilvopastoril, no noroeste de Minas Gerais. Foram coletadas amostras, nas profundidades de 0-5, 5-20 e 20-40 cm, de um Latossolo Vermelho Distrófico típico (LVd), entre os municípios de Vazante e Paracatu. Os sistemas foram selecionados segundo o histórico de uso: (1) CN- cerrado nativo (testemunha); (2) EA- eucalipto + arroz (ano zero do sistema agrossilvopastoril, plantados após o desmatamento da vegetação nativa de cerrado); (3) ES-eucalipto + soja (ano um do sistema, semeadura da soja em substituição ao arroz); (4) EP- eucalipto + pastagem (ano dois do sistema, com pastagem de Brachiaria brizantha Stapf., para a engorda do gado de corte); (5) EPG- eucalipto + pastagem + gado de corte (ano três do sistema, com a introdução do gado de corte); (6) PP- pastagem plantada de Brachiaria brizantha Stapf.; e (7) EC- eucalipto no sistema convencional no espaçamento de 2,0 x 3,0 m. O carbono orgânico e o carbono da biomassa microbiana apresentaram valores mais elevados na camada superficial (0-5cm), em relação às demais, em todos os sistemas. O carbono orgânico mostrou alteração substancial em relação aos sistemas estudados e às profundidades, revelando seu potencial como indicador da qualidade do solo em termos de reflexos das modificações impostas pelo manejo. Soil quality studies are important for understanding its interaction with the ecosystem as a whole. In this context, the soil microorganisms play a fundamental role for the maintenance and productivity of various agricultural ecosystems. This work had the objective to evaluate the biological attributes (microbial biomass carbon, basal respiration, and microbial quotient), as well as the organic matter content, possible indicators of alterations in soil quality imposed by different management systems, in relation to native cerrado, in agricultural - forestry - pasture system, in northwest of Minas Gerais state, Brazil. Samples were collected at 0-5, 5-20, and 20-40 cm depths, from a typical Dystrophic Red Latosol (Oxisol), between Vazante and Paracatu counties. The systems were selected according to the history of use, as follows: (1) CN - native cerrado (reference); (2) EA - eucalyptus + rice (year zero of agricultural - forestry - pasture system, planted after removal of cerrado native vegetation); (3) EP - eucalyptus + soybeans (year one of system, soybeans sowing substituting rice); (4) - EP - ES - eucalyptus + pasture (year two of system, using Brachiaria Brizantha Stapf. for cattle raising); (5) - EPG - eucalyptus + pasture + cattle (year three of system, with introduction of cattle); (6) PP - pasture cultivated with Brachiaria Brizantha Stapf.; and (7) EC - eucalyptus under conventional system in 2x3m spacing. The organic carbon and the microbial biomass carbon presented higher values in the superficial layer (0 - 5 cm), in relation to the other depths, in all systems. The organic carbon showed substantial alteration in relation to the studied systems and depths assessed, revealing its potential as soil quality indicator concerning the modifications imposed by management.
